LabonNet is developing Kineret, a software solution that will enable test of integrity of real-time PCR data together with data analysis of experimental assays.
Architecture Kineret has a modular architecture consisting of several analytical tools built on a hierarchical database. The power of Kineret emerges from integrating its database and novel functionality with existing and widely used analytical tools.
Work-flow An intuitive work-flow is facilitated in Kineret by passing data from one module to the next one, starting with the Outlier and ending up with the ERA. Reports from all computational modules are displayed in html in the Kineret's workshop and, in addition, a pdf report is produced.
Currently Kineret consists of following four modules:
Outlier - Outlier is a tool for detection and optional elimination of defective samples. The recognition is based on comparison of reaction kinetics as well as analysis of remote Ct values among group of replicated samples.
Calib - Inter-plate calibration of Ct values. Samples from various runs can be pooled and Ct values calibrated according to predefined calibrator samples. Large data sets can but thus analysed without limitation imposed by plate size.
RefFinder - Selection of optimal reference gene(s) based on pair-wise stability measure. This tool is an implementation of well known geNorm algorithm.
ERA - Expression Ratio Analysis facilitates relative quantification using reference genes with intelligent recognition of experimental design and selection of optimal statistical test. All types of replicates produced at any step of sample preparation are handled properly. Due to Kineret's unique tool for rapid experimental design definition, no knowledge of statistics is needed to obtain valid p-value for observed expression ratio.
